چکیده

background: in an effort to understand what limits the virulence of malaria parasites in relation to the host genetic and immunogenic background, we investigated the possibility that the parasite and host genotype crossover interactions constrain virulence. methods: two groups of mice from different genotypes were used (c57bl/6 (b6) and dba/2 mice). the mice were infected with a virulent parasite line plasmodium yoelii 17xl ( p. yoelii 17xl) . parasitemia, hematocrit value and lymphocytes yielded by livers and spleens were evaluated. fluorescence activated cell sorting (facs) analysis illustrated phenotypic characterization of lymphocytes. results: infection with p. yoelii 17xl did not result in thedeath of dba/2 mice. in contrast,b6 mice developed significantly high parasitemia and succumbed to death. using (facs) analysis, dba/2 mice were found to experience a marked expansion of interleukin (il)-2rb + cd3 int cells and gd t cells in the liver, especially in the recovery phase. the expansion of unconventional t cells (i.e. b220 + t cells) was also marked in dba/2 mice. conclusion: the outcome of murine malaria infections depends on the dynamic interplay between the immune-mediator and the genotype of the host.

Resistance and Susceptibility to Malarial Infection: A Host Defense Strategy against Malaria

BACKGROUND In an effort to understand what limits the virulence of malaria parasites in relation to the host genetic and immunogenic background, we investigated the possibility that the parasite and host genotype crossover interactions constrain virulence. METHODS Two groups of mice from different genotypes were used (C57BL/6 (B6) and DBA/2 mice). The mice were infected with a virulent parasi...

MyD88-Dependent Signaling Contributes to Host Defense against Ehrlichial Infection

The ehrlichiae are small gram-negative obligate intracellular bacteria in the family Anaplasmataceae. Ehrlichial infection in an accidental host may result in fatal diseases such as human monocytotropic ehrlichiosis, an emerging, tick-borne disease. Although the role of adaptive immune responses in the protection against ehrlichiosis has been well studied, the mechanism by which the innate immu...

Epithelial antimicrobial peptides in host defense against infection

One component of host defense at mucosal surfaces seems to be epithelium-derived antimicrobial peptides. Antimicrobial peptides are classified on the basis of their structure and amino acid motifs. Peptides of the defensin, cathelicidin, and histatin classes are found in humans. In the airways, alpha-defensins and the cathelicidin LL-37/hCAP-18 originate from neutrophils. beta-Defensins and LL-...

Role of inflammasomes in host defense against Citrobacter rodentium infection.

Citrobacter rodentium is an enteric bacterial pathogen of the mouse intestinal tract that triggers inflammatory responses resembling those of humans infected with enteropathogenic and enterohemorrhagic Escherichia coli. Inflammasome signaling is emerging as a central regulator of inflammatory and host responses to several pathogens, but the in vivo role of inflammasome signaling in host defense...

UV-B irradiation increases susceptibility of mice to malarial infection.

We here examined whether exposure of mice to UV-B affected their susceptibility to the murine malaria parasite Plasmodium chabaudi. When BALB/c mice with depilated skin were irradiated with UV-B and subsequently infected with the parasite, 80 to 100% of the UV-B-irradiated mice died within 12 days of infection with a sublethal dose. In addition, UV-B irradiation of C57BL/10 (B-10) mice, which a...

A new anti-malarial drug against murine malaria

Eosin B, a common laboratory dye has been earlier reported to have good anti protozoan properties in vitro. We studied this drug for its effect on a murine malaria strain, Plasmodium berghei (Pb) in vivo using different tests. Full suppressive 4 days Peter’s Test was used in infected outbred and inbred mice, using both ip and oral routes. Secondary biological assessment was carried out using do...
